RNA synthesis and uptake by liver cells in suspension

Abstract:

While it is generally agreed that organization of cells in tissue-specific patterns confers upon the cells certain characteristics which may not be obtained when the cells are not thus organized, there has been, so far, no systematic approach towards understanding the nature of those biochemical properties of cells in organized tissues which are a function of intercellular organization. One approach towards gaining useful information in regard to the biochemical basis of intercellular organization would be to compare the various biochemical properties of tissue slices with those of single-cell suspensions derived from the same tissue, and to see to what extent the biochemical changes obtained on dispersion of the tissue to a single cell suspension can be reversed on reaggregation of the cell suspension to tissue-like structures.1, 2

For the above studies it is necessary that the single cell suspensions-be obtained by a method which causes only those changes in the biochemical properties of the cells which are intrinsic to disruption of the intercellular organization and which are not caused by other physical or chemical-damage to the cells due to inadequacies of the method used for the dispersion of the tissue. We have recently described a method for the preparation of hepatic cell suspensions from rat liver which seems to satisfy the above criterion.3, 4
